Synthesis of copper protoporphyrin IX

Objective

Copper sulphate was reacted with protoporphyrin IX to produce copper protoporphyrin

Procedure

1. 25ml of DMF (solvent) was brought to boiling point under a condenser.
2. 0.2519g of protoporphyrin IX was added to the solvent and the mixture was allowed to boil for 20min.
3. 0.111g of copper sulphate was added to the boiling mixture and the reaction was allowed to run for 30min.
4. A dilute sample of the products was tested in the fluorescence spectrometry to see if the reaction has taken place or not.
5. Because the protoporphyrin was still emitting light at 640nm, it was concluded that the reaction was not complete, therefore 0.1254g of copper sulphate was added to the reaction to push the reaction toward the products.
6. After 20min, the reaction was still not complete therefore 0.163g of copper sulphate was added to the reaction again and the reaction was allowed to run for 20 more minutes.
7. After this period of time, the product was tested in fluorescence spectrometry again and because there was no peaks of light emission at 640nm, it was concluded that the reaction was complete.
8. The product of this reaction was added to 250ml of cold distilled water and was stored in refrigerator at 4 degree C.
