Objective

Description

  1. Prepare 10ml aqueous mixtures of BSA (1.5μM)and HAuCl4 (0.25mM) at room temperature.
  2. Prepare 10ml mixture of BSA (1.5μM)and HAuCl4 (0.25mM) using 50mM of Acetate as the buffer. The pH of the buffer was 4.3.
  3. Place each reaction mixture in an 80°C water bath.
  4. Obtain UV-Vis Spectra of gold colloidal suspensions in the wavelength range of 200-800nm at 30 minute intervals.

Data

2.5 mM of HAuCl4 and 15 μM of BSA were the two stock solutions used.

To prepare BSA (1.5μM)and HAuCl4 (0.25mM), the equation M1V1=M2V2 was used.

  • Calculation

2.5mM*V1 = 0.25mM*10ml

V1= 1ml of HAuCl4


0.015mM*V1 = 0.0015 mM*10ml

V1= 1ml of BSA


  • 1ml of HAuCl and 1ml of BSA were placed in each of the two test tubes.
