Objective

  • Test the effects of changing temperature on protein fiber formation.
  • In this experiment,the solution would be heated and then transferred into ice.

Description

Two solutions were prepared in eppendorf tubes.

  • Solution A which contains a molar ratio of HAuCl4 to BSA equal to 70 was prepared using the following procedure.
  1. 0.042mL of HAuCl4 (2.9mM), 0.1mL of BSA (17.7µM) and 0.85mL of H2O were pipetted into an eppendorf tube.
  2. The tube was placed in the 80°C heatblock for 1 hour.
  3. The tube was removed and placed in ice.
  • Solution B, which contains a molar ratio of HAuCl4 to BSA equal to 166 was prepared using the following procedure.
  1. 0.1mL of HAuCl4 (2.9mM,)0.1mL of BSA (17.7µM) and 0.8ml of H2O were pipetted into an eppendorf tube.
  2. The tube was placed in the 80°C heatblock for 1 hour.
  3. The tube was removed and placed in ice.

Data

  • Both Solutions A and B formed Purple Solution.
