Get the thermal cycler to 37 °C.
 
Once it is ready, get one of the 1 µL aliquots of 29% Rhodamine-Labeled tubulin suspension and place it on the thermal cycler for 30 min.
 
After 20 minutes have passed prepare 200 µL of Taxol in PEM solution. (198.8 µL of PEM and 0.2 µL of 10 mM Taxol in DMSO).
 
This last solution is added to 1 µL aliquots of 29% Rhodamine-Labeled tubulin suspension in the thermal cycler after the 30 minutes are up.
 
Finally, the polymerized microtubules need to be kept safe from ambient light.

Microtubule polymerization

  • This entry describes the steps for microtubule polymerization.

Procedure

Get the thermal cycler to 37 °C.
