*Each set consisted of 3 test tubes, each containing 4 stacked PLA scaffolds.
*All scaffolds were Autoclaved at then end of each procedure.

*100:1 Au:Myoglobin nanofibers were made based on previous biomineralization protocol
*The nanofibers were then place onto the scaffolds and allowed to dry

*100:1 Au:Myoglobin nanofibers were made following previously used protocol, but with the PLA scaffolds in the test tubes
 
===Polydopamine-coated scaffolds===

*Add 1mL of 0.0621 Myoglobin to each test tube, shake tube to combine, allow surface functionalization to occur for 5 hours
*Rinse with deionized water and allow scaffold to dry overnight
*Scaffolds submerged in water

Objective

  • Make 5 sets of PLA scaffolds: untreated PLA scaffolds, nanofiber-coated scaffolds by Brute-force method, biomineralization method of nanofiber formation with scaffolds, polydopamine-coated scaffolds, and polydopamine nanofiber-coated scaffolds
  • Collect result data from last lab

Methods

  • Each set consisted of 3 test tubes, each containing 4 stacked PLA scaffolds.
  • All scaffolds were Autoclaved at then end of each procedure.

Plain scaffold

  • The scaffolds were submerged in water

Brute Force method

  • 100:1 Au:Myoglobin nanofibers were made based on previous biomineralization protocol
  • The nanofibers were then place onto the scaffolds and allowed to dry
  • The dry nanofiber-coated scaffolds were then submerged in water

Old method + scaffolds

  • 100:1 Au:Myoglobin nanofibers were made following previously used protocol, but with the PLA scaffolds in the test tubes

Polydopamine-coated scaffolds

  • Scaffolds submerged in 10mM pH 8.46 TRIS dopamine hydrochloride solution for 1 hour
  • Add 1mL of 0.0621 Myoglobin to each test tube, shake tube to combine, allow surface functionalization to occur for 5 hours
  • Rinse with deionized water and allow scaffold to dry overnight
  • Scaffolds submerged in water

Polydopamine Nanofiber-coated scaffolds
