Objective

  • Perform gold nanoparticle(AuNP) synthesis using 17.7 μM Bovine Serum Albumin (BSA), 2.9 mM Chloroauric Acid (HAuCl4), and Water.
  • Test ratios of Au/BSA to better understand how to control purple protein fiber synthesis.
  • Two ratios specifically (70 and 166); according to previous results a 166 Au/BSA ratio yielded purple fibers and a ratio of 70 yielded purple solution.

Description

  • Created BSA Stock Solution (17.7µM): 0.0117g BSA, 1 mL Water.
  • Solutions A and B (Molar Ratio of Au/BSA = 70)

0.042 mL of HAuCl4 (2.9mM)

0.1 mL of BSA (17.7µM)

0.85 mL of water

Solution B was kept at constant 80 degrees celsius for 2 hours.

Solution A was removed from the heat block every 30 minutes for 10 minutes over the course of 2 hours.


  • Solutions C and D (Molar Ratio of Au/BSA = 166)

0.1 mL of HAuCl4 (2.9mM)

0.1 mL of BSA (17.7µM)

0.8 mL of water

Solution C was kept at constant 80 degrees celsius for 2 hours.

Solution D was removed from heat block every 30 minutes for 10 minutes over the course of 2 hours.

Data

  • Solutions A and B - Both were homogeneous purple solutions.
  • Solutions C and D - Both were clear solution with purple protein fibers.
  • Despite changes in temperature, an Au/BSA ratio of 70 yielded a purple solution , and a ratio of 166 yielded purple protein fibers.
  • Solutions from top to bottom: A, B, C, and D.


Notes

  • The results obtained from the experiment were consistent with results from previous experiments.
