Objective

Description

Part 1 : Preparation of porphyrin films

  • Add 2 drops of HCL, 1 ml of gluteraldehyde and 1 ml of porphyrin after the solution is homogeneous and cooled down.
  • Keep everything under the hood.

Part 2 : UV-VIS of porphyrin dilutions prepared on 17/10/2012

  • A) from the UV-Vis results we concluded that it is necessary to make extra 2 dilutions as dilution 1 and 2 are out of range. They are too concentrated. We would like to obtain a peak lower than 1.
  • Dilution 6: 4 ml of dilution 5 + 4 ml of HPLC water.(0.3375micM)
  • Dilution 7: 4ml of dilution 6 + 4 ml of HPLC water. (0.16785micM)

Part 3 : Prepare dilutions of 50micM Na2SO4, 50micM Al2(SO4)3, 50micM Fe2(SO4)3 (HPLC WATER IS USED)

Na2SO4

  • FW: 142.07 g/ml
  • 1/142.07 g/mol x 50micM/L x 10^(-6)mol/1micM x 1/1000ml x 10ml= 0.071035 g in 10 ml of HPLC water.
  • (50mM) (x) = (50micM) (10mL)
  • x = 0.01 ml =10micL of Na2SO4 in 9.99ml HPLC water


Al2(SO4)3

  • FW: 456.33g/mol
  • 456.33g/mol x 50mmol/L x 10^(-3)/1mmol x 1L/1000mL x 10 mml= 0.392g in 10 mL H2O.
  • 50micM=10micL of 50mM in 999micL H2O


Fe2(SO4)3 (72wt%)

  • FW:399.88g/mol
  • 0.72 x 399.88g/mol x 50mmol/L x 10^(-3)/1mmol x 1L/1000mL x 10 mml= 0.144g in 10 mL H2O.
  • 50micM=10micL of 50mM in 999micL H2O

Data

ISE results for sodium sulfate dilutions

  • Dilution 1(1.25ppm)= -180mV
  • Dilution 2(2.5ppm)= -182.6mV
  • Dilution 3(5ppm) = -182.4mV
  • Dilution 4(10ppm )= -177.4mV
  • Dilution 5(20ppm)= -174.6mV


  • We decided detection limit was between dilution 1 and 2.
  • More negative number means less sodium ion in the solution.
  • The porphyrin films prepared on 10/10/2012 had a yellowish green color. They were not clear as the other films.
