To 100ml of bacteria innoculum in a microvial, add 100ul of AMP and label as untreated | |||
To 100ml of bacteria innoculum in a microvial, add 100ul of AMP and 6ul of AMP and label as treated | |||
Put both microvial into the shaking incubate and leave for 15 mins | |||
Wash, centrifuge and vortex the microvials 3 times with 0.9% NaCl | |||
To the pellet after washing, add in 500ul of 50% ethanol and then vortex and centrifuge | |||
To the pellet after washing with 50% ethanol, add in 500ul of 90% ethanol and then vortex and centrifuge | |||
Pipette 10ul of each treated and the untreated mixture onto the microscope glass. Spread the samples beside each other | |||
Leave the sample to dry for 15 mins | |||
