Autoclaving PVA + Au:BSA Solutions

  • Using the 8 10mL test tubes created on Tuesday, add various substances and put all tubes into the autoclave.
    • Tube 1: 0.6g PVA 89-98K + 10mL water
      • add 1mL Gold + 1mL BSA
    • Tube 2: 0.6g PVA 130K + 10mL water
      • add 1mL Gold + 1mL BSA
    • Tube 3: 0.25mM Gold + 0.00147mM BSA + water
      • add 0.6g 89-98K PVA
    • Tube 4: 0.25mM Gold + 0.00278mM BSA + water
      • add 0.6g 89-98K PVA
    • Tube 5: 0.25mM Gold + 0.00147mM BSA + water
      • add nothing
    • Tube 6: 0.25mM Gold + 0.00278mM BSA + water
      • add nothing
    • Tube 7: nothing
      • add 1mL Gold + 1mL BSA + 8mL water
    • Tube 8: nothing
      • add 1mL Gold + 1mL BSA + 8mL water
  • All solutions put into the autoclave at 12:05PM

BSA and HAuCl4 Solutions

  • Au solution of HAuCl4:3H2O
    • MW = 393.83 g/mol
    • in 10mL 2.5mM solution, I need 9.096mg HAuCl4:3H2O
      • actual amount used: 10.21mg
      • therefore, I need 11.224mL H2O
  • BSA solution
    • MW = 66776g
    • in 20mL 15μM solution, I need 20.032mg BSA
      • actual amount used: 20.24mg BSA
      • therefore, I need 20.208mL of H2O
