[[User:Steven J. Koch|Steve Koch]] 18:41, 4 August 2009 (EDT): Made a thermal cycler program STEVE/ANNL, which hopefully will step temperature down from 95 to 4C in 0.01 ramp rate.

[[User:Steven J. Koch|Steve Koch]] 12:50, 5 August 2009 (EDT): Larry texted me last night to let me know it was down to 85.1 after about 10-15 minutes.  I came in this morning at it was holding at 4C.  So, I think it's safe to say that it slowly cooled down properly.  So, presumably I have some fork constructs now.

Steve Koch 18:27, 4 August 2009 (EDT): I want to see if the fork tether stuff from 2004 still works. It could be useful for a variety of things, including OT unzipping possibly. Found the stock oligos from AlphaDNA: http://openwetware.org/wiki/Koch_Lab:Protocols/Sequences/Unzipping_fork_oligos

Adding:

  • 10 microliters 3-DigFork1
  • 10 microliters 5-BioFork1
  • 0.64 microliters 3.9M NaCl
  • 5 microliters 1x TE
  • 25 microliters H2O
