Steve Koch 18:27, 4 August 2009 (EDT): I want to see if the fork tether stuff from 2004 still works. It could be useful for a variety of things, including OT unzipping possibly. Found the stock oligos from AlphaDNA: http://openwetware.org/wiki/Koch_Lab:Protocols/Sequences/Unzipping_fork_oligos

Adding:

  • 10 microliters 3-DigFork1
  • 10 microliters 5-BioFork1
  • 0.64 microliters 3.9M NaCl
  • 5 microliters 1x TE
  • 25 microliters H2O

Steve Koch 18:41, 4 August 2009 (EDT): Made a thermal cycler program STEVE/ANNL, which hopefully will step temperature down from 95 to 4C in 0.01 ramp rate.
