Fink et al 2001: "A new procedure for producing and purifying alpha -synuclein was developed by fusing its gene to a chitin-binding domain/intein system (IMPACT, New England Biolabs Inc.) and expressing the fusion protein in Escherichia coli. This system allowed simple purification of alpha -synuclein by binding of the chitin-binding domain fusion protein to a chitin column, followed by addition of a thiol agent (dithiothreitol or cysteine) to induce the cleavage reaction of the intein and to release the alpha -synuclein. The resultant protein, which was highly homogeneous upon polyacrylamide gel electrophoresis, was indistinguishable from authentic alpha -synuclein based on its electrophoretic mobility and molecular mass determined by electrospray mass spectrometry."
