Glad to see you thinking about this! Two things you should be aware of, and that will get you 95% of the way there. Maybe 99%:

  • Svoboda and Block review, something like "Application of optical trapping forces in biology..." --it's on the private wiki, in my references folder, etc. There's a section on calibration that explains the physics of the power spectrum.
  • Richard Yeh's master thesis. Also on the private wiki, and also available from Cornell's library (look for Richard's public OWW user page, I think he links it). This is really well written and explains calibration of our instrument (Tweezers #1) at Cornell.
