  • Ramalldf 20:03, 30 October 2009 (EDT):I made it a few months ago. Send it over, the reason that I used that font was cause I used the MASH font to make it look top secret for the private wiki logo so I just did the same but changed the colors.

Capping hairpin 2

Steve Koch 12:38, 11 August 2009 (EDT): OK, here's a google doc with my notes, and hopefully the same sequence as in the note below: {{#widget:Google Documents |key=0ARLNnjMk2r_qZGdxamtoNnBfMTE0ZDlmeDhjZjM |width=500 |height=300 }}

Capping hairpin

Here is the hairpin I used in grad school:

Unbiotinylated end-cap, HindIII

5’-P-TCGAGGCGACCTAGCGTAAGCTAGGTCGCC
Name = P16-Cap-HindIII(incorrect!!!!)
L=30
Purification = HPLC
Modifications = 5’ phosphate for ligation

I think you should:

  • Check with m-fold that it's a hairpin
  • replace the 3 or 4 base sticky end with 3 or 4 based for SapI that you have

GRD $$

Andy Maloney 11:16, 7 May 2009 (EDT): So Larry thinks I'm nagging about the GRD monies. I know I am but, I just cannot stop thinking about what we should get. And, since we are getting just about everything for the Kiney experiments really quickly, I think we should act on this sooner rather than later. I know you said you wanted me and Larry to tell you what you should get so here is what we came up with.

Water purifier

pH Meter

Update

Actually, I found a more all purpose meter from VWR.

It has software that comes with it and it comes with 3 probes for pH, conductivity, and dissolved oxygen content. Evans has an osmometer but hey, this may work for us. I'll get a quote for the stuff.

We will need to get pH standards for the meter and possibly conductivity standards but CRLS has those. I have a quote for the water filter stuff that is not listed on VWR's site so you are left with about $150 which should cover shipping charges for this stuff.

Can you let me know when you will be in the lab next? I think we should have a Koch Lab meeting soon.

Recombinant Casein Paper

Andy Maloney 16:42, 24 April 2009 (EDT): So this is the paper I found that talks about making E. coli produce cow utter puss. I can't understand any of it but if we do end up using [math]\displaystyle{ \kappa }[/math] casein a lot, we may want to make our own. If you are up for a challenge and read this, can you explain it to me?
