Hi John. The ratios are definitely crap, but even so, I have found this protocol very robust for generating PCR templates. In addition to regular PCR, I have used the DNA in quantitative PCR in a dilution series to calibrate different sets of primers and the standard curve was as tight as a dilution of cDNA. I have not used the DNA for anything else. Cheers, Michael |

DNA extraction
Hi John. The ratios are definitely crap, but even so, I have found this protocol very robust for generating PCR templates. In addition to regular PCR, I have used the DNA in quantitative PCR in a dilution series to calibrate different sets of primers and the standard curve was as tight as a dilution of cDNA. I have not used the DNA for anything else. Cheers, Michael
