pMAL-pIII is a plasmid that contains a gene for the periplasmic expression of Maltose Binding Protein (MBP). This plasmid is sold as a control plasmid to ensure that a lab's expression system is working properly. We are using it for the expressed purpose of producing MBP. We are using MBP to template the growth of bioconjugated gold nanoparticles.

pMAL expression and purification protocols

Of note:

  • Antibiotic: Ampcilin to 100μg/mL
  • Medium: 10g tryptone, 5g yeast extract + 2g glucose/L
  • IPTG stock (0.1M): 1.19g IPTG, 50mL water, filter to sterilize (cells are induced with 0.1mM IPTG)
