0.Preparation of Elementary part for two functions

   We made the DNA-conjugated gold nanoparticles which has two functions.


5’-thiol modified DNA attached to the gold nanoparticles



    We prepared 5’-thiol modified DNA and gold nanoparticles, and connect them each other by the specific binding between thiol group and gold nanoparticle. The specific binding between thiol group and gold nanoparticle is happen on theSelf-Assembled Monolayer(SAM). SAM is a monolayer which is built with the autonomous orientation by the interaction of gravitation. SAM is easily manufactured by putting substrates into a solution with a lot of molecules which have connectively functional group.    We confirmed this connection by hybridizing the complementary strand of 5’-thiol modified DNA with fluorophore. If 5’-thiol modified DNA is correctly attached to the gold nanoparticles, fluorophore binds to the gold nanoparticles through 5’-thiol modified DNA. So we can see whether 5’-thiol modified DNA are attached to the gold nanoparticles or not by measuring its fluorephore scence intensity spectrum.
   Figure 0-a, and c are fluorescence intensity spectrums from 490nm to 520nm. Figure0-a shows that there are two peaks, at 489nm and 520nm, and there are no peaks in b. The peak at 520nm indicates the fluorescence of fluorescence molecules. From these results, we concluded that the DNA binds to the gold nanoparticles.
