Summary Examined U2OS cells; found to be ~100%. Trypsinized cells with 2μL Trypsin. Incubated for ~3 minutes. Added 2ml cell culture media to wash off remaining cells from the flask. Added 3mL trypsinized cells in media to 21ml U2OS complete media. Pipetted up and down to mix. Added 9mL U2OS complete media to the remaining 1ml trypsinized cell suspension to split. Pipetted 4mL cell suspension in media to each well in 6 well plate. Placed split cell flask and 6 well plate in incubator.
