Aims For Friday

• Finish Stage three Ligations (With the exception of the aiiA part)

• Finish Stage two ligations (by wedensday)

• Test part T9002 (This part is the AHL/GFP assay and the Predator AHL sensor)

Aims For today

• Order Supplies John SY
• Maxiprep and electroporate for ligations Deepti, Farah, Johhny and John SY
• Test part T9002 Tom
• PCR Cre Lox part John C

Current Ligation Status

• Tick = Done and tested
• ? = Ligated and untested
• X = Done and Failed we will retry

Issues

Riboswitch

• Unable to tell if worked from PCR
• Will be sequenced (takes 2-3 days)

PCR

• The PCR results do not correlate with the miniprep results

minipreps

• These are not conclusive for many parts, we are looking into using a restriction digest
