Digestion test(again) Yesterday Daniel did the plasmid digestion again, but now with the correct plasmid. Today I did an electrophoresis 1% agar gel, 120 V x 35 min. Lanes: 1.Ladder 10 kb. 2.Digestion. 3.pBBRMCS-5. This shows that the plasmid was not digested because DH5-alfa cells are DCM+ so the next step is to transform the pBBRMCS-5 plasmid to other E. coli strain DCM- in this case BL21(DE3).
