Doing competent cells Daniel put 5 ml of liquid medium with a BL21's colony to grow since the morning. About 12:30 were picked up 100 micro-litters and passed into 1 litter. The cell grew up until reach OD = 0.1, Miguel said to us that because E. coli replicate every 30, we just needed to leave them growing 1 hour to reach the neccessary OD = 0.4 Once passed 1 hour, we centrifugated at maximum speed for 10 minutes and we descarted the supernatant. We added 1 ml of cool CaCl and vortex it, then we left them in ice all nigth. Tomorrow we'll finish the procedure.
