Grow Escherichia Coli DH5-α
Abstract
- Today I did three petri dishes with the bacteria Escherichia Coli DH5-α, which has the PBBMRCS-5 plasmid.
- Besides I did a agarose gel of the PBBMRCS-5 plasmid.
Grow E. coli DH5-α
- 1. Prepare LB medium.
- 2. Put 100 μl of gentamicin.
We do three tubes:
- 1. Control tube -> LB solid medium + gentamicin.
- 2. Bacteria tube -> LB solid medium + gentamicin + Escherichia coli DH5-α.
- 3. Bacteria tube -> LB solid medium + gentamicin + Escherichia coli DH5-α.
Note: The PBBRMCS-5 plasmid give to the bacteria resistance to gentamicin.
Gel of PBBMRCS-5 plasmid
- Line 1st -> 5 μl of Green ladder.
- Line 2nd -> 2.5 μl of plasmid + 2.5 μl of dye. (1)
- Line 3rd -> 2.5 μl of plasmid + 2.5 μl of dye. (2)
- Line 4th -> 5 μl of Green ladder.
Note: Previously the plasmid PBBMRCS-4 was digested with XBA I and Hind II.
