Description/Abstract

  • El principal objetivo del proyecto es hacer que la bacteria Rhizobium etli, simbionte del frijol y fijadora de nitrógeno, produzca Hidrógeno molecular en el ambiente apóxico que se genera en los nódulos de la raíz formados por la simbiosis entre Phaseolus vulgaris y Rhizobium etli.
  • The project's main objective is to produce molecular Hydrogen in Rhizobium etli. We'll take advantage of the apoxic enviroment generated during its symbiosis phase with the plant Phaseolus vulgaris because the enzymes capable of produce Hydrogen are sensitive to Oxygen, as some enzymes related with the nitrogen fixation process that takes place in R. etli.
  • El objetivo particular en el que yo me encuentro participando está relacionado con la creación de un kit para trabajar en Rhizobium etli. Dentro de este kit se pretende ofrecer una gama de promotores conocidos como promotores de la familia Anderson, se desea caracterizar el funcionamiento de estos promotores en Rhizobium etli.

General Workflow

  • Get DNA (20 promoters, J23100-J23119) from the iGEM distribution
  • Clone them on E. coli
  • Extract the plasmids and amplify the promoter part by PCR (Succesfully done on 03/09/2011)
  • Digest the amplified part with EcoRI and PstI (Done on 04/09/2011)
  • Introduce the promoter part (ligate) in plasmid pBBR1MCS5
  • Introduce the plasmid in E. coli S17 and then conjugate with R. etli.
  • Characterize the different promoters in R. etli.

  • I take this project in the amplification by PCR step
