July 6, 2010 Autoclaved glassware Made Terrific Broth without antibiotics Grew out CFP, YFP, and GFP Did a transformation of RFP that has a tetracycline resistance (RFP biobrick into TOP10 cells) Made solutions of copper solutions and TOP10 cells to determine optimal growth environment Analyzed alignment utilizing BLAST of construct templates and primer sequences Transformed YFP biobrick into TOP10 cells Grew out YFP, CFP, and YFP from glycerol stock for testing of fluorescence using the plate reader in the morning Contacted team about next regional meeting Discussed skype calls and established set individuals on each team that would be in charge of maintaining communication over skype
