08.07.2009 Compotent cell culture was taken from incubation. Process of growing competent cell continued. 50 ml. LB was put in 3 flasks. 300µl bacteria solution which is waited in incubation for 14- 16 hours is put in each flask. Again incubation at 320 rpm for 2-2.5 hours. After 2 hours we measured OD (600 nm) with spectrophotometer. result = 0.24 So we continued incubation because we want result of OD is about 0.6 After 1.5 hours, we measured again. Our results ; 1.flask = 0.67 2.flask = 0.71 3.flask = 0.74 we used first flask and kept on process. 10 mM and 75 mM CaCl2 was prepared. for 10 mM = 0.044g CaCl2 + 40 ml dH2O for 75 mM = 0.8325g CaCl2 + 10 ml dH2O ampicilline was made. 10 ml dH2O + 1 gr. Amp LB Agar was prepared.
