Talk:Knight:M9 supplemented media
I noticed on the wiki that Knight lab uses 0.2% casamino acids in supplemented M9 media. Is this for faster growth?? I use 0.1%, and I think this is what most people in the Endy lab use.
- I looked into M9 media recipes a while back and there are recipes calling for anywhere between 0.1%-0.5% casamino acids. For instance, Ron Weiss (and Barry who obtained Ron's recipe) use 0.2%. Jen and Caitlin's recipe calls for 0.1%. The Microbiological Media Handbook suggests 0.5%. The Difco manual's recipe calls for 0.4%. I basically just used 0.2% as a compromise. It is hopefully high enough to provide plenty of amino acids for the cells but low enough that there shouldn't be too much in the way of trace amounts of sugars that can play havoc with inducible promoters (like lactose and the Plac promoter). --Reshma 18:12, 25 May 2005 (EDT)
