So it looks like the picture isn't much better, but the noise is reduced and some artifacts have been removed. I would say SapI definitely cut, NotI likely, and it looks like ligation to SapCap produces some concatamers so maybe increasing the concentration of that would be useful. I think later I will ligate my SapCap to the anchor construct, and then plan out digestion overnight with SapI of Clone14 for direct ligation to the tethering construct. Maybe as 2 ligations? I'll speak with Koch on this and get back to myself.
