The Daily Grind

  • So I have decided I will grow all my varieties here and start them all out in a growth room in small pots before transplant to the nursery. An article I read suggests Mexican plants at the 3 leaf stage need 14 days of 10 hours light/ 14 hours dark and they will be programmed to flower. This would most conveniently be done in a growth room, although it could also be done with trash cans in the field. Here's the article:
  • New recipe for Salkowski reagent : From Gordon and Weber (1951) (GW reagent). Mix sample 1:2 with reagent which consists of 1 ml 0.5 M ferric chloride plus 50 ml 35% (w/w) perchloric acid. Will have dilutions standardized from 500, 400, 300, 250, 125, 62, 31, 15, 7.5, 4, 2 ug/ mL. Let reaction proceed for 30 minutes. This was from Appl Microbiol Biotechnol (1997) 48: 726-729
  • Above reaction just barely worked - doesn't appear to be sensitive enough (see photos). Standards were 100 ug IAA in 100 ul, diluted in half all the way to 0.7 g. Also included were two lanes with either 10 ug or none of IAA with 100 ul E. coli in each. Same done for tryptophan (starting at 100 ul of 0.5 M stuff) and 2,3 Butanediol starting at 2 ug in 200 ul. (this assay needs to be done in a ratio of 2:1 sample to alpha-napthol)
