The Daily Grind Replating soil endophytes out using glycerol stocks. Taking 10 ul of glycerol (800 ul of juice plus 500 ul of 80% glycerol) and resuspending in 290 ul of phosphate buffer (plus cycolhexamide). Spreading 50 ul of that dilution on R2A plates. Cut potatoes for BAC survival study. Plated ligations of AraBad, Medium Promoter, and Weak Promoter to pSB1A2
