The Daily Grind To include the spermosphere in the analysis as a potential source of bacteria, 5 seeds of each genotype was placed in 5 ml of the buffer/tween overnight at room temperature in a lightly closed 15 ml falcon tube. After 24 hours, seeds will be shaken, removed from the tube, and the buffer frozen for later DNA extraction. Gonna have to use the Ultraclean DNA extraction kit from MoBio for some samples, cause 100 preps is not enough...
