Objective

To observe pepsin's ability to cleave hemoglobin with and without pepstatin.

Procedure

The procedure was followed according to Prof. Harting's instructions, with a few changes.

  1. The concentration of pepstatin in the parallel solution was 200nM
  2. The stock pepsin solution was diluted from 12μM to 2nM using .83μL in 5mL total solution
  3. The perchloric acid was 1M, not 1.7M
  4. The hemoglobin stock solution was diluted (10μL to 1mL) and used as a UV-Vis reference

  • UV-VIS spectra data, taken at 1/2 hour intervals
