Objective

  • Fluorescence of varying concentrations of dye in distilled water / dye in buffer
  • Testing fluorescence of the BSA+DYE+buffer
  • test fluorescence of blank (buffer)
  • Concentration Curve

Description

Stock Solutions:

  • 50µM of dye in phosphate buffer
  • 0.001 grams of dye + 50 ml buffer
  • 50 µM of dye in distilled water
  • 0.001 grams of dye + 50 ml buffer


Varying Concentrations of Dye – Concentration curve

  • Add 200µL of stock to 800µL of buffer = 10 µM
  • Add 100µL of stock to 900µL of buffer = 5 µM
  • Add 20µL stock to 980 µL of buffer = 2 µM

Resulting Concentrations

  • 1µM, 2µM, 5µM, and 10µM
  • 13µM(30/1000) = .381 µM of dye in the dye+BSA

Notes

  • it appears to be a shift for the BSA + dye
