05/19/2010: Repeating luciferase purification... FAILED Today, I started the day by repeating the DNA purification from a gel band using low melting point agarose 2%. After electrophoresis at 95V for 1 hour,I purified the band as previously done. Looking forward to observing the purified luciferase, I ran another gel; however... the band was not there!!! Tomorrow, I will ask Miguel for advice to run a PCR, as the purification will probably not give any output as far as the previous results indicate. I also made petri plates, 13 of LB with ampicilin and 12 of LB with kanamycin.
