Repeating the transformation Today, I made the transformation of the ligation realized yesterday. The resulting strains were plated in LB agar and left to grow for 8 hrs. The plates with their respective strains are: 1. Ligation of luciferase and double terminator (T)in a LB with kanamycin plate. 2. The ligation control (plasmid 18 already cut with ) in a LB with tetracycline plate. 3. The transformation control (pSB1T3 with an insert of RFP) in a LB with tetracycline plate. 4. Control of competent cells in a LB with tetracycline plate.
