Objective

Acquire fluorescence scans of myoglobin at pH 4 and pH 5 at 80C.

pH 4 fluorescence

3 mL Sample

  1. 1 μM myoglobin
    1. Stock Concentration 160 μM
    2. Volume Added: 18.75 μL
  2. pH 4
    1. Stock Concentration 1 mM HCl
    2. Volume Added: 300 μL
  3. Volume water added: 2681 μL

UV-Vis of sample

Definite unfolding of myoglobin as heme is misligated.

Fluorescence scans

  1. Acquisition information
    1. Excitation: 290 nm, 10 nm slit width
    2. Emission: 310-540 nm, 10 nm slit width
    3. 100 nm/minute
    4. 61 measurements, 180 second delay between measurements


pH 5 Fluorescence

3 mL Sample

  1. 1 μM myoglobin
    1. Stock Concentration 160 μM
    2. Volume Added: 18.75 μL
  2. pH 5
    1. Stock Concentration 1 mM HCl
    2. Volume Added: 30 μL
  3. Volume water added: 2961 μL

The initial scan for the pH4 looks much different than the initial scan for the pH 5
