Entry title

1.Beer's Law standard curve

1. 50 mL of a BSA stock solution that is roughly 5 mg in 5 mL of saline was prepared by adding 50mg solid BSA to 50ml if saline solution.

2. UV-VIS spectra was recorded between 200 nm and 800 nm for both stock solution and saline solution( see figure 1).

3. 6 standard solutions (1 mL each) were made in concentrations of 1µg/ml, 3µg/ml, 5µg/ml, 10µg/ml, 15 µg/ml, and 20µg/ml (table 1)

  • 200 μL of 4:1 diluted Bio-Rad Protein Assay reagent were added to each tube
  • the correct amount of Tris/NaCl buffers added such that the final volume is 1 mL.
  • tubes were closed and vortexed for 5 - 10 sec.
  • Left to sit for 5 min.

4.UV-VIS spectrum was obtained and record between 400 nm and 800 nm 5. duplicate blanks (4 solutions total) were made:

  • 1 mL Tris/NaCl buffer
  • 200 μL Bradford reagent + 800 μL buffer
