Objective

  • Perform gold nanoparticle(AuNP) synthesis to obtain purple homogeneous solution using 15.5 μM Bovine Serum Albumin (BSA), 2.9 mM Chloroauric Acid (HAuCl4), H2O, and Tris.

Description

  • Prepared four tubes:

Tube 1: 0.1 mL BSA, 0.1 mL HAuCl4, 0.8 mL H2O.

Tube 2: 0.1 mL BSA, 0.1 mL HAuCl4, 0.8 mL Tris Buffer (pH 9.36).

Tube 3: 0.1 mL BSA, 0.05 mL HAuCl4, 0.85 mL H2O.

Tube 4: 0.1 mL BSA, 0.05 mL HAuCl4, 0.85 mL Tris Buffer (pH 9.36).

  • Tubes placed in heat block set at 80 degrees celsius, for 2 hours.
  • Heat block was turned off after 2 hours to facilitate gradual decrease in temperature.

Data

  • Tube 1: Purple protein fiber aggregation, Molar ratio of Au/BSA = 187
  • Tube 2: Clear solution, Molar ratio of Au/BSA = 187
  • Tube 3: Purple solution (similar to experiment performed on 9/28), Molar ratio of Au/BSA = 98.4
  • Tube 4: Clear solution, Molar ratio of Au/BSA = 98.4
