Ran gel of yesterday's PCR
- 1hr 20 min @ 120V
- Somewhat inconclusive results, but 5.1 is definitely positive. Will most likely confirm via cross to Flp-out.
Observed tdt-Myb, gfp-PCNA
- Too much background in tdt-Myb channel
Dissected tdt-Myb, gfp-PCNA
- Orange and Peach/Yellow eyes
- Fix with 4% Formaldehyde PBS Solution
  - 20 minutes
- Wash 3 x 3 minutes in 1x PBS-T
- Incubate 25 minutes with PBS 0.6% Triton.
- Wash once with 1x PBS-T
- Block O/N 1:1000 in 2% NGS
