Miniprep, ran plasmid on gel Performed a miniprep of the DNA extracted from E. coli, expecting to obtain as plasmid with insert+backbone Miniprep/Qiagen_kit Performed a restriction digest on the plasmid with EcoRI and PstI then denatured the restriction enzymes. Ran on gel, found backbone without an insert (single piece of DNA 2.6kb in length). Verified original plasmid had mutation added (removal of XbaI site in ORF).
