Objective Analyze the intein reactions that were set-up yesterday. Description Take fluorescence spectra of the MBP-intein reactions with the gold nanoparticles from yesterday (tubes 7-12). Also take fluorescence spectra of unreacted (mixed in cuvette, and then immediately placed in the fluorometer) MBP-intein and gold nanoparticle mixtures. Data For the graphs where the "not reacted" data line is not easily seen, it follows the same line as "reacted at 37°C" Notes There is a major problem with getting protein gels (SDS-page) to polymerize for use. It would have been helpful to have seen the reaction mixtures on a gel.
