CNHY / CNTI Multiplex & SCOC MPY sar80

Pour Load Run
270 mL 1x TAE + 5.4 g agarose 2 µL gel load dye + 4 µL PCR product 120 V
27 µL GelRed 6 µL ladder 25 minutes


  • only used 1 row; saved rest of gel for later use


  • this is just confirmation gel, so not all samples were run on gel
  • for CNHY / CNTI gel lanes, used CNHY062911TKLH
  • Uniplex: A1-7
  • Multiplex all-in-one: A8-9
  • Multiplex A & B: G1, G4
  • for SCOC gel lanes, used first three
  • G7-9 (to include water)


Gel - SCOC MPY sar80 trial

  • water is clean
  • two bands visible, in range of Scun3 (200bp) and sar80 (300bp) (but Scun11 band also in 300bp range)
  • hoping all three amplicons are visible in frag analysis


Gel - CNHY / CNTI Uniplexes

  • some uniplexes didn't seem to amplify
  • not gunna worry about it, since amplification successful in previous uniplex (2012 January 9 Uniplex gel)
  • assuming other samples in this PCR had successful amplification


Gel - CNHY / CNTI Multiplex all-in-one

  • only see one prominent band in the 200bp range
  • very strong primer banding (inefficient PCR)  :(


Gel - CNHY / CNTI Multiplex A and Multiplex B

  • no strong bands, only faint banding
  • bands do seem to be different between MPA and MPB
  • very strong primer banding (inefficient PCR)  :(


  • good enough - send to frag!
