Objective

To observe the catalytic activity of pepsin (in cleaving peptide bonds in hemoglobin) in the presence and absence of pepstatin. This data will be compared with data we take in a later lab on pepsin-AuNPs.

Procedure

Preparation of Pepsin-Hemoglobin Sample

  1. Add 0.83 μL of pepsin to 4.99 ml of hemoglobin solution, creating a final concentration of 2 nM.
  2. Incubate the tube at 37°C for 30 minutes.
  3. Take aliquots every 30 minutes for 2 hours.

Preparation of Pepstatin-Hemoglobin Sample

  1. Add 0.83 μL of pepstatin to 4.99 ml of hemoglobin solution, creating a final concentration of 200 nM (0.2 μM).
  2. Incubate the tube at 37°C for 30 minutes.
  3. Take aliquots every 30 minutes for 2 hours.
