Objective

  • Test solvents and ionic strength on all ratios of AuNPs.
  • Take 1 mL aliquots of AuNPs made yesterday.
  • Take notes on the effect of chelators and pH.
  • Test the effect of SDS.
  • Test the effect of pepsin.

Procedure

Solvent Tests

  • 0.5 ml of DMF, DMSO, chloroform, 1,4 dioxane, and acetone were added to a 1 ml aliquot of each ratio AuNP.
  • Tubes were vortexed and allowed to sit overnight.
  • Note: no immediate change was noticed.

Ionic Strength

  • Bulk amounts of NaCl, KCl, MgCl2, and CaCl2 were added to each ratio.
  • Tubes were vortexed and allowed to sit overnight.
  • Note: no immediate change was noticed.

Random Tests

  • 0.1 g of SDS was added to each ratio.
  • A bulk amount of pepsin was added to a single tube of 50:1 AuNPs.
  • Note: no immediate change was noticed.

Observations

  • The addition of chelators and pH change did not cause the fibers to fall apart.
  • 2,2 biyridine induced fiber formation in the 40:1 tubes.
  • More acidic solutions induced fiber formation in the 40:1 tubes. Basic pH did not have an effect.
  • There was a clear difference between associating and non-associating bases with pH.

Notes

  • The 40 and 50 samples from yesterday both made fibers.
