Objective
- Test solvents and ionic strength on all ratios of AuNPs.
- Take 1 mL aliquots of AuNPs made yesterday.
- Take notes on the effect of chelators and pH.
- Test the effect of SDS.
- Test the effect of pepsin.
Procedure
Solvent Tests
- 0.5 ml of DMF, DMSO, chloroform, 1,4 dioxane, and acetone were added to a 1 ml aliquot of each ratio AuNP.
- Tubes were vortexed and allowed to sit overnight.
- Note: no immediate change was noticed.
Ionic Strength
- Bulk amounts of NaCl, KCl, MgCl2, and CaCl2 were added to each ratio.
- Tubes were vortexed and allowed to sit overnight.
- Note: no immediate change was noticed.
Random Tests
- 0.1 g of SDS was added to each ratio.
- A bulk amount of pepsin was added to a single tube of 50:1 AuNPs.
- Note: no immediate change was noticed.
Observations
- The addition of chelators and pH change did not cause the fibers to fall apart.
- 2,2 biyridine induced fiber formation in the 40:1 tubes.
- More acidic solutions induced fiber formation in the 40:1 tubes. Basic pH did not have an effect.
- There was a clear difference between associating and non-associating bases with pH.
Notes
- The 40 and 50 samples from yesterday both made fibers.
