Objective

To lower the pH of the dye reactions (to stop the reactions) and to perform dialysis on our samples.

Description

  1. Lower the pH of the BSA/HCl control with dye to 6.59 by adding 1μL of 1 M HCl. The solution became pink.
  2. Lower the pH of the 166 molar ratio with dye to 5.53 by adding 37 μL of 1 M HCl. The solution became light pink.
  3. Lower the pH of the 70 molar ratio with dye to 6.83 by adding 5 μL of 1 M HCl. The solution became purple.
  4. The solutions were transferred to Fisherbrand regenerated cellulose dialysis tubing. (flat width = 19 mm, vol/cm = 1.15 mL, wall thickness = 25μm, and dry cylinder diameter = 12.1 mm)
  5. The dialysis tubing containing the solutions were placed in a dialysis container filled with water and covered with aluminum foil.
  6. Over the course of the following week, the water was changed 7 times.
