* Extracting DNA from 2010 distribution.  
*Plate 1. 7A. Plasmid pSB1T3 with 10μL H<sub>2</sub>O millinQ
***Transform DH5α competent cells.
***1.-Cells defrosted on ice.
***2.-100μL of cells for 3μL of DNA from plate.
***3.-3 tubes were generated and transformed. 1 control with 50μL of cells.
***4.-20 minutes on ice.
***5.-Thermal shock at 42°C, 1 minute.
***6.-5 minutes on ice.
***7.-Add 1 ml of LB at each tube.
***8.-1 hour at 37°C.
***9.-Plate on Tet30-LB medium
***Odd cells stored at 4°C.

WetLab

  • Extracting DNA from 2010 distribution.
  • Plate 1. 7A. Plasmid pSB1T3 with 10μL H2O millinQ
      • Transform DH5α competent cells.
      • 1.-Cells defrosted on ice.
      • 2.-100μL of cells for 3μL of DNA from plate.
      • 3.-3 tubes were generated and transformed. 1 control with 50μL of cells.
      • 4.-20 minutes on ice.
      • 5.-Thermal shock at 42°C, 1 minute.
      • 6.-5 minutes on ice.
      • 7.-Add 1 ml of LB at each tube.
