09/06/17 Observations from last class It can be seen that pink particles were formed in the isoleucine solutions as well as in the 1:1:1 1M phenyl solution. blue coloring indicates beginning of NP formation. '''1. Titrant standerization''' - 5mM NaOH solution was prepared from 1M stock solution using M1V1=M2V2 - since the stock solution was prepared from solid NaOH, the solid absorbed some water while in the container and therefor 2ml of tock solution were added preparation. -A buret was rinsed with about 10 mL of NaOH solution, emptied completely, and washed and drained again - 0.001 g of KHPhth were transferred to an Erlenmeyer flask with 30 mL of water. - A few drops of phenolphthalein solution were added - Titrated until the solution turns pink. Repeated twice. 1st trial: 29ml 2nd trial : 25.7 ml - 10 mL of 250 μM amino acid solution was transferred to a clean and dry 100 mL beaker - 10 mL of 200 μM HCl and 10 mL water were added to the beaker - I pH probe was inserted to solution and initial values recorded - titrant was added in 2 - 4 mL intervals. pH recorded after gentle swirling -Continued until the pH reaches at least 10 or 11. -was done for Isoleucine and Phenylalanine - need to be done for arginine Figure 2: Amino acids titration volumes and graphs NK
