Objective

Determine the Au/Gold ratio at which the solution becomes fibers

Description

Created a gradient with increasing AU/BSA ratios.

Gold stock- 2.9mM BSA stock 17.7μM

Up to this point we have found that at low Au/BSA ratios a solution is formed, and at high Au/BSA ratios fibers are formed. Today we are running a gradient to determine where and how that change occurs.

Tube Number 1 2 3 4 5 6
Amount of 2.9mM HAuCl4 100 μL 120 μL 150 μL 180 μL 200 μL 250 μL
Amount of 17.7μM BSA 100 μL 100 μL 100 μL 100 μL 100 μL 100 μL
Amount of Distilled Water 800 μL 780 μL 750 μL 720 μL 700 μL 650 μL
Ratio [Au]/[BSA] 163.842 196.91 245.763 294.915 327.684 409.605

Left in heatblock at 80°C for approximately 3 hours.

When the tubes were removed from the heatblock, the test tube 1 and 2 initially appeared to be a purple solution. Within a minute purple fibers came out of solution and the solution became clear. The other test tubes appeared as shown below. There was a small golden aggregation in test tubes 3-6.
