Date: 30 January 2009; Time: 1000; Agenda: Mid Log Phase Determination
- 1 loop of PA01 colony inoculated in 150ml LB Broth at 37 degrees incubator, 200rpm.
- For the first 3 hrs, the absorbance is recorded every 30min. Subsequently, the absorbance at 600nm was recorded hourly until 12 hrs is reached. A final 24 hr is recorded the next day.
- Tare spectrophtometer with pure LB Broth first before recording.
This time we did not plate the samples.
