08/30/2018-Experimenting with Aliphatic Amino Acids and NaOH

Only Methionine showed a strong shift, though it turned black- no gold nanoparticles but at least it was reactive. This could be because of the sulfur in the R-group. From this we planned a pH assay with both Methionine and Cyestine (which also has a sulfur).

For an acidic component: HCl Stock HCl = 36.5% solution --> halved --> 18.25 % --> 9.125% solution --> 2mL stock solution/ 6mL DI water

  • 36.46 g/mol --> MW of HCl

pH Assay: Acidic-

  • 5mM HCl

Control- no pH modifiers Basic-

  • 1.25 mM NaOH
  • 2.5 mM NaOH
  • 5 mM NaOH
  • 7.5 mM NaOH

Two assays: one using Meth one using Cys

  • 2mL Gold solution
  • 2mL Base/Acid
  • 2mL Amino Acid

4 hours in an 80℃ oven- samples were looked at the following Tuesday (9/11/18) Biggest Changes: Meth 5mM NaOH- deep red Cys 5mM NaOH- Grey/blue precipitated out
