Purpose

The purpose of this assignment is to explore the biological mechanisms behind the genetics of flowers, specifically in their expression of color as we are setting out to create a true-breeding purple plant in the Aipotu software using two green, one red, and one white flowers.

Methods/Results

Part 1

  1. Self crossed each of the four given flowers (Green-1, Green-2, Red, White)
  2. Crossed each of the given flowers with one another.
  3. Performed back cross to gain understanding of dominance between alleles.
  4. Recorded all the alleles found along with their associated color.
  5. Performed experiment using Aipotu software in order to create a purple flower.

Genotypes of Starting Strains

  • CrCr = white
  • CGCG = green-1
  • CRCr = red
  • CBCY = green-2

Identification of Alleles

  • Cr = White
  • CR = Red
  • CG = Green
  • CB = Blue
  • CY = Yellow

Identification of Possible Genotypes

  • CBCB = Blue
  • CBCY = Green
  • CGCR = Black
  • CRCY = Orange
  • CRCB = Purple

Dominance of Alles: {CG ≈ CR; CR = CB = CY; CG > CB = CY > Cr}

  • ≈ is incompletely dominant
  • = is equally dominant
  • > is dominant

Purple is formed due to the incomplete dominance when crossing CBCB & CRCR and getting CBCR. In this case, it was a 50% chance of producing the purple flower as anticipated using Mendelian genetics (1:2:1).

Part 2

  • Created Homozygous pure-breeding blue flower by performing multiple crosses and a self-cross
  • Used Aipotu mutation function to mutate flower
  • Compared the mutant and pure breeding flower and recorded the discrepancies in protein and DNA sequences

Conclusion

In this lab, we set out to create a pure breeding purple strain. Unfortunately, even after many trials of mutating and breeding, we were unable to form a pure-breeding purple plant. We assumed this to be the case due to the incomplete dominance between the CB and CR alleles in the genotype which resulted in its purple color in the first place. Because there is no allele specific to the purple color, it is impossible to create a pure-breeding purple flower.
